It marked Best's second career round win, with the Victorian finishing second on Saturday.
The Tickford Racing driver leaving the first round of a new season as points leader for the second time in as many seasons.
Matt Payne and pole man Declan Fraser finished second and third respectively.
MW Motorsport teammates Thomas Maxwell and Tyler Everingham were fourth and fifth.
Fraser and Triple Eight teammate Cameron Hill led through Turn 1 as front row starter Angelo Mouzouris bogged down.
Mouzouris sent Hill into a spin at Turn 2 and was forced into retirement with damage.
It wasn't the only incident on the opening lap; a start-line incident between James Masterton, Paul Boschert and Tony Auddino triggered the first Safety Car period.
The race restarted with Fraser in the lead, but a daring move at Turn 5 helped Best steal the front running.
Payne followed through into second, with Fraser losing two positions in a matter of corners.
The Safety Car made its second appearance when Zane Morse and Cameron Crick locked horns exiting Turn 8.
Crick slid across the wet grass and gently nudged the concrete, and was unable to extract his Eggleston Commodore.
Crick's teammate Matt McLean, already carrying rear damage, was the centre of an incident which ended the races of Ryal Harris and Nash Morris.
At the lap 11 restart, McLean ran Harris wide onto the grass; an out-of-control Harris slid across the infield an took out an unsuspecting Morris.
A third Safety Car deployment ensured the race ran time certain, handing Best just his second career Super2 win.
Harris and Morris were left trackside remonstrating over the incident, both drivers making their full Super2 debuts this weekend.
Jaylyn Robotham, Saturday's winner, finished sixth ahead of Aaron Seton, Matt Chahda and Jay Hanson, with McLean classified 10th.
Kai Allen scored Super3 honours in 11th overall.
The 2022 Dunlop Series will resume in Perth next month.
